Title: The Effects of Biomembrane “Defects” on Lipid Transfer Rate and Hydrophobic-Hydrophilic Interactions

Abstract:

Phospholipids are amphiphilic molecules and the building block of biological membranes. The interactions between lipids and membrane-associated biological molecules, e.g., proteins, glycolipids, cholesterol, etc. have attracted enormous attentions because they control the activities on the biomembranes. There are hundreds of natural or synthetic phospholipids which differ in regards to the properties of hydrophilic headgroups (i.e., charge density and size) and hydrophobic acyl chains (e.g., number, length and level of saturation of the hydrocarbons). These differences vary the molecular spontaneous curvature, hydration, miscibility, solubility and crystallinity of the lipid molecules, resulting in segregation of lipids on the bilayer and/or formation of special morphology. One of the special scenarios is “bicelle” which is a mixture of long- and short- chain lipids forming well-defined nanodiscs with a bilayer thickness and uniform radius. Bicelle showed much higher cellular uptake than liposomes, the most common drug delivery carriers, and thereby has great potential to be applied for the future delivery carriers. In this talk, I will provide some fundamental understanding about how the defects on the bilayer induced by the lipid segregation influence the lipid exchange kinetics and lipid interactions with other amphiphilic molecules, hoping to provide the insight into the activities associated with biomembranes.
