ABSTRACT
Solution crystallization is a notoriously complicated process where the final crystal product is a function of thermodynamics as well as kinetics, physics and transport phenomena. The results of the crystallization step have far reaching impacts on overall process efficiency, final product quality and reproducibility such as purity, yield, polymorphism, stability, particle size distribution, filterability, flowability, drying ability and dissolution rate. The simple translation of a description of a laboratory synthetic process into a series of sequential unit operations required to replicate the procedure in process equipment is complicated. Materials balance and real-time monitoring of crystallization are important and shown in this presentation to provide many benefits leading to the comprehension of crystallization theory, improved methods for process development, optimization, scale-up and the compilation of log sheet. A batch wise production of acetaminophen will be given as an example.
